Hypervitaminosis A and Collagen Synthesis

On average every year as we get older our serum retinol increases, our liver retinol stores increase and our rate of collagen synthesis decreases. Could there be a connection? Studies into the effect of Hypervitaminosis A on bone reveal that there is likely a direct connection. It appears that Hypervitaminosis A weakens bone due to decreasing collagen synthesis in bone. Why wouldn't it then decrease collagen synthesis everywhere in the body and why would the progressive loss of collagen synthesis with age not be partly due to vA bioaccumulation?

Effects of Hypervitaminosis A on the Bone and Mineral Metabolism of the Rat

"Vitamin A toxicity has been associated with alterations in mineral metabolism and may result in osteopenia, fractures, deformities, and growth arrest. The pathogenesis of the bone lesions that occur in vitamin A toxicity is, however, ill defined and was examined in the present study. The administration of pharmacological doses of vitamin A to growing male rats resulted in weakness and spontaneous fractures. Undecalcified bone histology of vitamin A toxic animals was characterized by increased bone resorption, osteoclastosis, a paucity of trabecular surfaces covered with osteoid, and lesions which appear to be pathognomonic of hypervitaminosis A. The serum calcium and magnesium levels of vitamin A-toxic animals were unremarkable, but serum phosphate levels were significantly higher than control values. Urinary hydroxyproline excretion reflected bone histology and was significantly increased in experimental rats. Circulating levels of the potent bone resorbers, PTH, 1,25-dihydroxyvitamin D, and 25-hydroxyvitamin D, were, however, comparable in vitamin A-toxic and control animals, suggesting a possible direct effect of vitamin A on bone. Subsequently, the effects of vitamin A (retinol) on in vitro collagen synthesis (incorporation of [3H]proline into collagen) and bone resorption (45Ca release from bone) were examined using a fetal rat calvarial culture. Retinol added to the culture medium for 20-24 h in concentrations ranging from 0.5-10 micrograms/ml selectively inhibited collagen synthesis in a dose-dependent fashion. Higher concentrations of retinol were toxic and resulted in a general inhibition of protein synthesis. Bone resorption was stimulated by 0.5 and 2.5 micrograms/ml retinol. We conclude that vitamin A toxicity in rats causes bone lesions, the genesis of which can be explained, at least in part, by a direct effect of the vitamin on skeletal tissue."

It is thought that increasing the amount of Vit C and gelatin in the diet helps to boost collagen synthesis:

Vitamin C–enriched gelatin supplementation before intermittent activity augments collagen synthesis

"Results: Supplementation with increasing amounts of gelatin increased circulating glycine, proline, hydroxyproline, and hydroxylysine, peaking 1 h after the supplement was given. Engineered ligaments treated for 6 d with serum from samples collected before or 1 h after subjects consumed a placebo or 5 or 15 g gelatin showed increased collagen content and improved mechanics. Subjects who took 15 g gelatin 1 h before exercise showed double the amino-terminal propeptide of collagen I in their blood, indicating increased collagen synthesis."

How much does this really help though? Young people usually have great collagen synthesis despite consuming low levels of gelatin and Vit C and there are many older people that consume plenty of both that have poor collagen synthesis.

I've spoken at length about my grandmother, her naturally lowish VA diet, her longevity.   Even in her late 90's my grandmother's bones were wide and sturdy.  At the very end of her life she started to show some ridges in her nails and brought them to my attention.  Others here have posted about nails.

I would bet that some crooked teeth are down to this collagen thing.  Price was probably close to right-- that you need to get the fat soluble activators IN CORRECT AMOUNTS for proper growth of the face.  Not that you had to have LOADS, but they have to be CORRECT.   (whatever that is for YOU)

My grandmother was so very good with her VA status, but things went south with my mother her daughter, and on to me.  I sort of wonder if there was problematic synthetic VA in the formula she fed my mother, that started a downward spiral. 

The blueprint for tooth growth begins to work right in the womb.  My mother has straight teeth, but she is the last in her line with them.  Mine are crooked, as are my children's.

Crooked teeth are caused by small jaws.

One cause of lack of mid facial development is head trauma, the biggest source being the birthing process. Babies should always be taken to an experienced cranial osteopath asap after birth.

Pacifiers are another massive cause of jaw growth disturbances.

Children should always have something to chew as well, some kids like to chew on bones (never cooked bones as they can splinter).

Hypervitaminosis A in early life likely plays a major role. Vit D deficiency is a major player with regard to bone growth abnormalities. Vitamin K2 status is very important. There are probably many other nutrients and other causes that can play a role too. Even barefoot walking probably promotes better cranial development...

or carrots like my 3yo niece...

I find my mother's case interesting.  I'm pretty sure she was baked in a womb of a non-VA poisoned person, but that shortly afterwards she began to be one herself.  (she had failure to thrive).  Straight teeth.

Then, I was (possibly) baked in a VA-toxic womb, and my kids too.  Crooked teeth.
